The great dane had originally a connection with german mastiffs. I do not know about the modern ones that seem to live and average of 8 years and are really friendly.
Most mastiffs originally had long heads. The shorter heads were developed for the various bulldog breeds which were basically catch dogs. They were designed to grab and not let go. Fighting dogs grab and rip. Slightly longer jaws are better for creating more drastic wounds. Wild canines like wolves have longer jaws and kill and scavenge for their food. The majority of mastiff breeds sold today are modern recreations invented for the dog show ring and also to sell pups. Their prime objective is to make them real big. Behavior and health is secondary. For some reason they have decided to give them short and and often lower undershot jaws. The teeth instead of being long are often short. More suitable for a catch dog than a war dog that kills.
